Step-by-Step Home DNA Collection: Buccal Swab Instructions & Prep

How to Collect Your DNA Sample at Home: Step-by-Step Buccal Swab Instructions

Our at-home DNA testing kit makes sample collection easy and accurate. Follow these important steps for the best results:

  • Wait at least 30 minutes after eating, drinking, or smoking before collecting your sample.

  • Complete all paperwork before you begin the process.

  • Have your sample envelope ready and within reach.

  • Carefully review your kit instructions to avoid common mistakes.

For most tests, use the included cheek (buccal) swab for DNA collection.

Note: For non-invasive prenatal paternity (NIPP) testing, a blood sample from a licensed provider is required. Cheek swabs are not sufficient for NIPP, except for the alleged father only.

For detailed, visual guides, refer to your kit materials, the guide below or contact our support team if you have questions. Proper preparation ensures fast, accurate results and a smooth testing experience.

Step 1: Prepare for DNA Collection

Wait 30 minutes after eating, drinking, or smoking to ensure a clean DNA sample. Avoid brushing teeth or using mouthwash immediately before swabbing.

Step 2: Remove the Swab Carefully

Open the swab packaging without touching the fabric tip to any surface. Only handle the swab by the stick for best sample quality.

Step 3: Swab Cheeks Thoroughly (30 Seconds Each Side)

Firmly roll the swab against the inside of your cheek for 30 seconds on each side. Target the cheek, not the gums, to collect the most DNA-rich cells.

Step 4: Air Dry the Swab (60 Seconds)

Wave the swab in the air for one minute if it appears wet. The swab does not need to be completely dry before packaging.

Frequently Asked Questions about Home DNA Test Collection & Buccal Swab Kits

  • Swab each cheek firmly for at least 30 seconds as instructed. Use all provided swabs, alternating cheeks, and avoid contact with teeth or gums for the best DNA cell collection.

  • Use a new swab if available. Touching the fabric tip to fingers, counters, or other surfaces may contaminate your sample and affect test results.

  • Only use the swabs supplied in your kit. Regular cotton swabs aren’t designed for DNA preservation and may not be accepted by the lab.

  • Reschedule the collection and wait 30 minutes after last eating, drinking, or smoking to avoid contamination.

  • Use another kit swab if available. If you need a replacement, contact GameDay DNA for a new sample collection kit.

  • Request a new envelope or dry thoroughly before mailing. Wet paper can degrade DNA or cause delays in processing.

  • Using mouthwash or taking medication before swabbing may interfere with DNA cell recovery. Follow guidelines—wait 30 minutes after mouthwash or medication before collection.

  • No—always place swabs into the provided paper sample envelope. Plastic can create moisture and degrade the DNA sample.

  • Paper-packed cheek swabs are viable for testing for several weeks if kept dry and at room temperature. Mail your samples promptly after collection.

  • Follow kit instructions strictly and label each sample. For multi-person tests (paternity, siblings, grandparents), use a separate swab and envelope for each individual.
